The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Taste Receptor Type 2 Member 38 (TAS2R38). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Taste Receptor Type 2 Member 38 (TAS2R38). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Taste Receptor Type 2 Member 38 (TAS2R38), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Taste Receptor Type 2 Member 38 (TAS2R38) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a seven-transmembrane G protein-coupled receptor that controls the ability to taste glucosinolates, a family of bitter-tasting compounds found in plants of the Brassica sp. Synthetic compounds phenylthiocarbamide (PTC) and 6-n-propylthiouracil (PROP) have been identified as ligands for this receptor and have been used to test the genetic diversity of this gene. Although several allelic forms of this gene have been identified worldwide, there are two predominant common forms (taster and non-taster) found outside of Africa. These alleles differ at three nucleotide positions resulting in amino acid changes in the protein (A49P, A262V, and V296I) with the amino acid combination PAV identifying the taster variant (and AVI identifying the non-taster variant). 1. We now describe expression of T2R38 in tumor cells in patients with pancreatic cancer and in tumor-derived cell lines. our data indicate a new, additional function of the taste receptor T2R38 beyond sensing "bitter".
