Myeloid ecotropoic viral insertion site 1 (MEIS1) is a transcription factor belonging to TALE (three amino acids loop extension) family. The gene for MEIS1 is mapped to human chromosome 2p15. The amino terminus contains binding domains for its interaction partners, the transcription factors HOXA9 and PBX1, followed by a homeobox DNA binding domain and carboxy-terminal transcriptional activation domains. It is expressed in hematopoietic stem and progenitor cells and increases during human megakaryocytic differentiation. MEIS1 plays a role in the normal development of the hematopoietic lineage and is over expressed in a subset of acute myeloid leukemias. Increased MEIS1 expression has also been observed in neuroblastomas and the expression level of the MEIS1 transcript is a prognostic indicator in breast cancer.

MEIS1: Acts as a transcriptional regulator of PAX6. Acts as a transcriptional activator of PF4 in complex with PBX1 or PBX2. Required for hematopoiesis, megakaryocyte lineage development and vascular patterning. May function as a cofactor for HOXA7 and HOXA9 in the induction of myeloid leukemias. Defects in MEIS1 could be a cause of susceptibility to restless legs syndrome type 7 (RLS7). Restless legs syndrome (RLS) is a neurologic sleep/wake disorder characterized by uncomfortable and unpleasant sensations in the legs that appear at rest, usually at night, inducing an irresistible desire to move the legs. The disorder results in nocturnal insomnia and chronic sleep deprivation. Belongs to the TALE/MEIS homeobox family. 2 isoforms of the human protein are produced by alternative splicing.
