MBS285177 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the 1,25-Dihydroxyvitamin D3 (1,25 DHVD3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing 1,25 DHVD3. The ELISA analytical biochemical technique of the MBS285177 kit is based on 1,25 DHVD3 antibody-1,25 DHVD3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect 1,25 DHVD3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, 1,25 DHVD3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

1,25-(OH)2D3, the active metabolite of vitamin D, regulates immune responses in addition to its role in calcium, phosphorus, and bone metabolism. Recent data suggests that 1,25-(OH)2D3 blocks dendritic cell maturation influencing the development of regulatory T cells. High dose 1,25-(OH)2D3 monotherapy is effective at delaying acute rejection. The vitamin D receptor (VDR), which functions as a transcription factor, forms a complex with Smad 3, a TGFb-1 signaling protein, in rat renal lysates for treated recipients. 1,25-(OH)2D3 significantly prolonged graft survival, limited the degree of intersitial fibrosis and glomerulosclerosis, decreased urinary protein and altered Smad and MMP expression in a rat renal model of CAN. In a retrospective clinical study, that cadaveric renal transplant recipients with renal insufficiency placed on calcitriol demonstrate improved renal function and improved graft survival.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate 25-Dihydroxyvitamin in samples. An antibody specific for 25-Dihydroxyvitamin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any25-Dihydroxyvitamin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for 25-Dihydroxyvitamin is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 25-Dihydroxyvitamin bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for 1,25 DHVD3
This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This mitochondrial protein initiates the degradation of 1,25-dihydroxyvitamin D3, the physiologically active form of vitamin D3, by hydroxylation of the side chain. In regulating the level of vitamin D3, this enzyme plays a role in calcium homeostasis and the vitamin D endocrine system. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
UniProt Comments for 1,25 DHVD3
CYP24A1: Has a role in maintaining calcium homeostasis. Catalyzes the NADPH-dependent 24-hydroxylation of calcidiol (25- hydroxyvitamin D(3)) and calcitriol (1-alpha,25-dihydroxyvitamin D(3)). The enzyme can perform up to 6 rounds of hydroxylation of calcitriol leading to calcitroic acid. It also shows 23- hydroxylating activity leading to 1-alpha,25-dihydroxyvitamin D(3)-26,23-lactone as end product. Defects in CYP24A1 are the cause of hypercalcemia infantile (HCAI). HCAI is a disorder characterized by abnormally high level of calcium in the blood, failure to thrive, vomiting, dehydration, and nephrocalcinosis. Belongs to the cytochrome P450 family. 2 isoforms of the human protein are produced by alternative splicing.
