Biological Significance
BRCA 1 and 2 are breast cancer susceptibility proteins that play a role in DNA damage, repair, cell cycle progression, transcription and apoptosis, etc. The BRCA 1 gene is expressed in numerous tissues, including breast and ovary, and encodes a predicted protein of 1863 amino acids. BRCA2 has been shown to be required for localization of Rad51 to sites of DSBs (double stranded breaks) in DNA. Cells lacking BRCA 1 and BRCA 2 cannot repair DSBs through the Rad51 dependent process.
Immunogen
BRCA 2 antibody was raised in Rabbit using synthetic peptide mapping to the N-terminus of human BRCA2 as the immunogen.
Preparation and Storage
Store at -20 degree C. For long-term storage, aliquot and freeze at -70 degree C. Avoid repeated freeze/thaw cycles.

If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, Dec 2008]
