The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing COL17A1. The ELISA analytical biochemical technique of the MBS945372 kit is based on COL17A1 antibody-COL17A1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect COL17A1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, COL17A1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for COL17A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any COL17A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for COL17A1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of COL17A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes the alpha chain of type XVII collagen. Unlike most collagens, collagen XVII is a transmembrane protein. Collagen XVII is a structural component of hemidesmosomes, multiprotein complexes at the dermal-epidermal basement membrane zone that mediate adhesion of keratinocytes to the underlying membrane. Mutations in this gene are associated with both generalized atrophic benign and junctional epidermolysis bullosa. Two homotrimeric forms of type XVII collagen exist. The full length form is the transmembrane protein. A soluble form, referred to as either ectodomain or LAD-1, is generated by proteolytic processing of the full length form. Defects in COL17A1 are a cause of generalized atrophic benign epidermolysis bullosa (GABEB).
