The ELISA analytical biochemical technique of the MBS2705324 kit is based on ADRa2C antibody-ADRa2C antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ADRa2C antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ADRa2C. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Adrenergic Receptor Alpha 2C (ADRa2C). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Adrenergic Receptor Alpha 2C (ADRa2C). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Adrenergic Receptor Alpha 2C (ADRa2C), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Adrenergic Receptor Alpha 2C (ADRa2C) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Alpha-2-adrenergic receptors are members of the G protein-coupled receptor superfamily. They include 3 highly homologous subtypes: alpha2A, alpha2B, and alpha2C. These receptors have a critical role in regulating neurotransmitter release from sympathetic nerves and from adrenergic neurons in the central nervous system. The mouse studies revealed that both the alpha2A and alpha2C subtypes were required for normal presynaptic control of transmitter release from sympathetic nerves in the heart and from central noradrenergic neurons. The alpha2A subtype inhibited transmitter release at high stimulation frequencies, whereas the alpha2C subtype modulated neurotransmission at lower levels of nerve activity.
