Description: This assay employs a two-site sandwich ELISA to quantitate OTC in samples. An antibody specific for OTC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any OTC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for OTC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of OTC bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Ornithine transcarbamoylase (OTC) (also called ornithine carbamoyltransferase) is an enzyme that catalyzes the reaction between carbamoyl phosphate (CP) and ornithine (Orn) to form citrulline (Cit) and phosphate (Pi). In plants and microbes OTC is involved in arginine (Arg) biosynthesis, whereas in mammals it is located in the mitochondria and is part of the urea cycle.OTC is a trimer.
The monomer unit has a CP-binding domain and an amino acid-binding domain. Each of the two discrete substrate-binding domains (SBDs) have an alpha/beta; topology with a central beta-pleated sheet embedded in flanking alpha-helices.The active sites are located at the interface between the protein monomers.

NCBI Summary for OTC
This nuclear gene encodes a mitochondrial matrix enzyme. Missense, nonsense, and frameshift mutations in this enzyme lead to ornithine transcarbamylase deficiency, which causes hyperammonemia. Since the gene for this enzyme maps close to that for Duchenne muscular dystrophy, it may play a role in that disease also. [provided by RefSeq, Jul 2008]
