Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

PTF1A (Pancreas transcription factor 1a; also p48 and bHLHA29) is a cell specific member of the bHLH family of proteins. Although its predicted MW is 41kD, a high ratio of Pro, Leu and Ala increases its MW to 48kD in SDS-PAGE. PTF1A forms a trimeric transcription factor complex with a p75 subunit (HEB, E2 or E47) and a p64 subunit (usually RBP-L). Within this complex, unfortunately termed PTF1, p48/PTF1A binds to a DNA E-box while p64 binds to a TC-box. This regulates the gene transcription of about 20 enzymes synthesized by pancreatic acinar cells. Human PTF1A is 328aa in length. It contains one basic DNA binding HLH motif (aa150-175) and a dimerization interface (aa178-219). Overaa1-92, human PTF1A shares 88% aa identity with mouse PTF1A.

This gene encodes a protein that is a component of the pancreas transcription factor 1 complex (PTF1) and is known to have a role in mammalian pancreatic development. The protein plays a role in determining whether cells allocated to the pancreatic buds continue towards pancreatic organogenesis or revert back to duodenal fates. The protein is thought to be involved in the maintenance of exocrine pancreas-specific gene expression including elastase 1 and amylase. Mutations in this gene cause cerebellar agenesis and loss of expression is seen in ductal type pancreas cancers. [provided by RefSeq]
