Description: The protein encoded by this gene is a member of the G-protein coupled receptor family 2. This protein is a receptor for parathyroid hormone (PTH) and for parathyroid hormone-like hormone (PTHLH). The activity of this receptor is mediated by G proteins which activate adenylyl cyclase and also a phosphatidylinositol-calcium second messenger system. Defects in this receptor are known to be the cause of Jansen's metaphysealchondrodysplasia (JMC), chondrodysplasiaBlomstrand type (BOCD), as well as enchodromatosis. Subunit Structure: Interacts (via N-terminal extracellular domain) with PTHLH and PTH (PubMed:8397094, PubMed:10913300, PubMed:18375760, PubMed:19674967). Homodimer in the absence of bound ligand. Peptide hormone binding leads to dissociation of the homodimer (PubMed:19674967, PubMed:20172855).

Immunofluorescene (IF) of anti-PTH1R antibody
MBS9607245 staining Hela by IF/ICC. The sample were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25 degree C. The primary antibody was diluted at 1/200 and incubated with the sample for 1 hour at 37 degree C. An Alexa Fluor 594 conjugated goat anti-rabbit IgG (H+L) Ab, diluted at 1/600, was used as the secondary antibody.

Immunohistochemistry (IHC) of anti-PTH1R antibody
MBS9607245 at 1/100 staining Human liver cancer tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the antibody for 1.5 hours at 22 degree C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.
