Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team

Principle of the Assay: AT1R-AA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AT1R-AA antibody and an AT1R-AA-HRP conjugate. The assay sample and buffer are incubated together with AT1R-AA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AT1R-AA concentration since AT1R-AA from samples and AT1R-AA-HRP conjugate compete for the anti-AT1R-AA antibody . Since the number of sites is limited, as more sites are occupied by AT1R-AA from the sample, fewer sites are left to bind AT1R-AA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AT1R-AA concentration in each sample is interpolated from this standard curve.

Angiotensin II is a potent vasopressor hormone and a primary regulator of aldosterone secretion. It is an important effector controlling blood pressure and volume in the cardiovascular system. It acts through at least two types of receptors. This gene encodes the type 1 receptor which is thought to mediate the major cardiovascular effects of angiotensin II. This gene may play a role in the generation of reperfusion arrhythmias following restoration of blood flow to ischemic or infarcted myocardium. It was previously thought that a related gene, denoted as AGTR1B, existed; however, it is now believed that there is only one type 1 receptor gene in humans.
