MBS2022672 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Transaldolase (TALDO1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TALDO1. The ELISA analytical biochemical technique of the MBS2022672 kit is based on TALDO1 antibody-TALDO1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TALDO1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TALDO1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Transaldolase (TALDO1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Transaldolase (TALDO1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Transaldolase (TALDO1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Transaldolase (TALDO1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Transaldolase 1 is a key enzyme of the nonoxidative pentose phosphate pathway providing ribose-5-phosphate for nucleic acid synthesis and NADPH for lipid biosynthesis. This pathway can also maintain glutathione at a reduced state and thus protect sulfhydryl groups and cellular integrity from oxygen radicals. The functional gene of transaldolase 1 is located on chromosome 11 and a pseudogene is identified on chromosome 1 but there are conflicting map locations. The second and third exon of this gene were developed by insertion of a retrotransposable element. This gene is thought to be involved in multiple sclerosis. [provided by RefSeq, Jul 2008]

TALDO1: Transaldolase is important for the balance of metabolites in the pentose-phosphate pathway. Defects in TALDO1 are the cause of transaldolase 1 deficiency (TALDO1 deficiency). It results in telangiectases of the skin, hepatosplenomegaly, and enlarged clitoris. Belongs to the transaldolase family. Type 1 subfamily.
