anti-CD9 antibody
CD9 antigen is a protein that in humans is encoded by the CD9 gene.1 CD9 was found to be intensely expressed on the cell surface of the glandular epithelium throughout the menstrual cycle without any apparent differences in staining intensity. In addition, Western blotting analysis of the affinity-purified proteins confirmed that CD9 was associated with integrins beta 1, alpha 3 and alpha 6 in the human endometrium. CD9 may be an important regulator of these integrins in the human endometrium.2 This protein also seems to be a key part in the egg-sperm fusion during mammalian fertilization, as CD9 knocked-out mice gametes don't undergo fusion.3 CD9 is located in the microvillar membrane of the oocytes and also appears to intervene in maintaining the normal shape of oocyte microvilli.4

NCBI Summary for CD9
This gene encodes a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Tetraspanins are cell surface glycoproteins with four transmembrane domains that form multimeric complexes with other cell surface proteins. The encoded protein functions in many cellular processes including differentiation, adhesion, and signal transduction, and expression of this gene plays a critical role in the suppression of cancer cell motility and metastasis. UniProt Comments for CD9
Function: Involved in platelet activation and aggregation. Regulates paranodal junction formation. Involved in cell adhesion, cell motility and tumor metastasis. Required for sperm-egg fusion. Ref.11 Ref.12 Ref.13
Subunit structure: Forms both disulfide-linked homodimers and higher homooligomers as well as heterooligomers with other members of the tetraspanin family. Associates with CR2/CD21 and with PTGFRN/CD9P1. Interacts directly with IGSF8. Ref.14 Ref.16 Ref.17
Subcellular location: Membrane; Multi-pass membrane protein.
Tissue specificity: Expressed by a variety of hematopoietic and epithelial cells.
Post-translational modification: Protein exists in three forms with molecular masses between 22 and 27 kDa, and is known to carry covalently linked fatty acids.
Sequence similarities: Belongs to the tetraspanin (TM4SF) family.

1. Data indicate that CD9 acts as scaffold and assembles a ternary JAM-A-CD9-alphavbeta3 integrin complex from which JAM-A is released upon bFGF stimulation.
