The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing F13B. The ELISA analytical biochemical technique of the MBS752304 kit is based on F13B antibody-F13B antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect F13B antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, F13B. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: F13B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-F13B antibody and an F13B-HRP conjugate. The assay sample and buffer are incubated together with F13B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the F13B concentration since F13B from samples and F13B-HRP conjugate compete for the anti-F13B antibody . Since the number of sites is limited, as more sites are occupied by F13B from the sample, fewer sites are left to bind F13B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The F13B concentration in each sample is interpolated from this standard curve.

This gene encodes coagulation factor XIII B subunit. Coagulation factor XIII is the last zymogen to become activated in the blood coagulation cascade. Plasma factor XIII is a heterotetramer composed of 2 A subunits and 2 B subunits. The A subunits have catalytic function, and the B subunits do not have enzymatic activity and may serve as a plasma carrier molecules. Platelet factor XIII is comprised only of 2 A subunits, which are identical to those of plasma origin. Upon activation by the cleavage of the activation peptide by thrombin and in the presence of calcium ion, the plasma factor XIII dissociates its B subunits and yields the same active enzyme, factor XIIIa, as platelet factor XIII. This enzyme acts as a transglutaminase to catalyze the formation of gamma-glutamyl-epsilon-lysine crosslinking between fibrin molecules, thus stabilizing the fibrin clot. Factor XIII deficiency is classified into two categories: type I deficiency, characterized by the lack of both the A and B subunits; and type II deficiency, characterized by the lack of the A subunit alone. F13B: The B chain of factor XIII is not catalytically active, but is thought to stabilize the A subunits and regulate the rate of transglutaminase formation by thrombin. Defects in F13B are the cause of factor XIII subunit B deficiency (FA13BD). FA13BD is an autosomal recessive disorder characterized by a life-long bleeding tendency, impaired wound healing and spontaneous abortion in affected women.
