Ribosomal S6 Kinase 2 is an 83kD member of the S6 kinase subfamily of Ser/Thr protein kinases. The kinase is downstream of the extracellular signal-related kinase (ERK)/MAPK cascade and mediates growth-factor and stress induced activation of the transcription factor cAMP response element-binding protein. Ribosomal S6 Kinase 2 has been shown to inhibit G protein-coupled receptors and it's loss leads to Coffin-Lowry syndrome and nonsyndromic X-linked mental retardation. It is widely expressed, primarily in lung and skeletal muscle.

The activity of this protein has been implicated in controlling cell growth and differentiation. Mutations in this gene have been associated with Coffin-Lowry syndrome (CLS).
