Description |
The decidual cells play an important role in the maintenance of pregnancy at the maternofetal interface after implantation of the fertilized egg into the endometrium. The Immortalized Rat Decidual Cells (GG-AD) is conditionally immortalized through the temperature-sensitive simian virus (SV40 tsA209) and possesses several endocrinological features, including the expression of progesterone receptor, activin type II, activin βA and activin βB. In addition, it also retains the ability to respond to cAMP stimulation and secrete PRL-related hormones, a hallmark of the antimesometrial decidual cells. This cell line is useful for studying the mechanism of decidual cell regression and reorganization. |

Propagation |
Use of PriCoat? T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow II medium available from ABM (TM002). To make the complete growth medium, add the following components to the base medium: 10% fetal bovine serum (TM999) and Penicillin/streptomycin. Atmosphere: air: 95%, CO2: 5%; Temperature: 33.0°C. The functional expression of the SV40 antigen is induced by culturing the cells in vitro at a temperature permissive (33°C). At a non-permissive temperature (37°C-39°C), the cells cease to proliferate and revert to differentiated phenotype. |
Preservation |
1. Freeze Medium: Complete growth medium with 20% FBS and 10% DMSO.
2. Storage Temperature: Liquid nitrogen vapour phase. |
Quality Control |
1) Northern blot performed to analyze presence of activin βA , follistatin and PLP-B mRNA; 2) RT-PCR analysis of progesterone receptor, activin βA, activin βB, and activin receptor II mRNA was completed with L19 ribosomal protein mRNA as control. |
