Complement convadase assay是一種用于確定生物材料補(bǔ)體激活的靈敏且特異性的方法。
在與血漿孵育過程中,補(bǔ)體因子與材料表面結(jié)合,然后形成補(bǔ)體合酶復(fù)合物。 洗滌后,將生物材料在含有特定顯色底物的培養(yǎng)基中孵育,以去除未結(jié)合的補(bǔ)體蛋白。 通過測量 405 nm 的光密度來量化底物裂開。

◆ 原理
將感興趣的測試物質(zhì)(理想情況下應(yīng)知道表面積)作為血漿孵育。 在孵育過程中,補(bǔ)體因子與材料表面結(jié)合,然后形成補(bǔ)體自相酶復(fù)合物。
孵育后,洗滌樣品并使用補(bǔ)體相容酶特異性顯色底物分析補(bǔ)體相容酶活性。 顯色速率取決于生物材料表面產(chǎn)生的相聚酶的量。

產(chǎn)品在 -20°C 下可穩(wěn)定保存至少 6 周
